Imprimeur-libraire ; architypographe du Roi [d'Espagne]. - Vers 1557, il entre au service de l'imprimeur-libraire d'Anvers Christophe Plantin à qui il succédera en 1589 et dont il épouse la fille Martine en 1570. Travaille à Venise chez Girolamo Scoto entre 1562 et 1565 lors de la fermeture de l'officine de Plantin, alors accusé d'hérésie. Admis dans la guilde de Saint-Luc d'Anvers en qualité de maître imprimeur en août 1586, il obtient patente royale en fév. 1587. C. Plantin lui lègue son imprimerie d'Anvers, son matériel et son fonds de librairie, au grand dam des héritiers, avec qui Joannes I Moretus doit trouver un compromis. A traduit en néerlandais le "De constantia" de Juste Lipse et le "Premier Jour" de la "Première Semaine de la Création du monde" de Guillaume Du Bartas

Moretus werd officiële stadsdrukker van Antwerpen (tot 1705); Jean I Moretus verkreeg monopolie als officiële drukker van verschillende religieuze orden (zoals Spaanse jezuïeten t.t.v. de Contrareformatie)

Drukarz i księgarz (Antwerpia - Officina Plantiniana). Działał w latach 1586-1610. W 1589 objął drukarnię po śmierci swego teścia Ch. Plantina. Od przejęcia przez niego oficyny do śmierci Jeanne Rivière, wdowy po Krzysztofie Plantinie, firma działała pod nazwą: "Ex Officina Plantiniana apud Viduam et Ioannem Moretum" (1590-1596), następnie prowadzili ją dwaj jego synowie: Joannes Moretus II (1576-1618) i Balthasar Moretus (1574-1641).
